AOH1996是复制子异构体成分PCNA(增殖细胞核抗原)的口服活性配体,靶向转录-复制冲突(TRC)。AOH1996稳定PCNA和RNA聚合酶II的相互作用,导致rpb1的蛋白酶体依赖性降解和致命的DNA损伤。AOH1996还干扰PCNA及其结合蛋白的相互作用,导致DNA复制应激并诱导细胞凋亡。AOH1996与DNA损伤剂协同作用,抑制肿瘤细胞[1][2]。
